促红细胞生成素对脑创伤后细胞凋亡和Caspase-3活性的影响

Effect of recombinant human erythropoietin on nervous cell apoptosis and activity of Caspase-3 after brain injury in rats

摘要 目的探讨重组人促红细胞生成素(rhEPO)对大鼠创伤性脑损伤的治疗作用及对细胞凋亡和Caspase-3活性的影响之间的关系。方法制作SD大鼠自由落体脑挫裂伤模型,设立假手术组、rhEPO治疗组和对照组,分别应用透射电镜、流式细胞技术和荧光四肽底物法检测各组大鼠脑挫伤灶旁细胞凋亡情况及Caspase-3活性。结果脑外伤后存在细胞凋亡,细胞凋亡的发生在伤后24h达到高峰,rhEPO治疗组在伤后24h和48h细胞凋亡发生率低于相应的对照组(P〈0.05);rhEPO治疗后12,24和48h大鼠脑挫伤灶旁细胞Caspase-3活性均低于各自时间点对照组(P〈0.05或0.01)。结论rhEPO治疗对创伤性脑损伤后细胞凋亡的发生及Caspase-3的活性有影响,抑制细胞凋亡及Caspase-3活性下降很可能是rhEPO对创伤性脑损伤治疗作用的重要机制。 Objective To investigate the relationship between the therapeutic effect of recombinant human erythropoietin (rhEPO) on focal brain contusion in rats and its impact on apoptosis and the activity of Caspase-3. Methods The rats were randomly assigned three groups : the control, trauma and treatment with rhEPO groups. Focal brain contusion was induced in the latter two groups. Apoptosis of neuron cells and the activity of Caspase-3 was evaluated with transmission electron microscopy, flowcytometry and AC-DEVD-AMC. Results At 24 hours in the cortex and 48 hours in the hippocampus, apoptosis rates of neuronal cells for the reEPO treatment group were significantly lower than those of the control ( P 〈 0. 05 ). Activity of Caspase-3 ( P 〈 0. 05 ) was reduced markedly in rats treated with rhEPO. Conclusion rhEPO is closely correlated with apoptosis and the activity of Caspase-3 after traumatic brain injury, and apoptosis inhibition and decreased activity of Caspase-3 by rhEPO may be an important mechanism for treatment of focal brain contusion after traumatic brain injury.
