摘要
目的:观察清开灵不同组分对MCAO大鼠脑组织肿瘤坏死因子-α(TNF-α)及可溶性肿瘤坏死因子受体(sTNF-R)含量的影响,探讨其抗脑缺血损伤的机制。方法:采用MCAO模型,应用RIA检测脑组织匀浆肿瘤坏死因子-α(TNF-α含量,ELSIA检测脑组织sTNF-R含量。结果:缺血24h,胆酸组、栀子苷组、珍珠母组及合方组的脑组织中TNF-α含量显著降低(与模型组比P<0.05或P<0.01)。缺血12h,珍珠母组、黄芩苷组和胆酸组脑组织中sTNF-R的含量显著高于模型组(P<0.05或P<0.01);而缺血24h,胆酸组与合方组脑组织中sTNF-R的含量显著低于模型组(P<0.05或P<0.01)。结论:清开灵四种组分及合方可以抑制TNF-α的合成与分泌,对脑缺血后的级联反应具有阻抑作用;而各不同组分对sTNF-R的作用较为复杂,在缺血12h,珍珠母、黄芩苷和胆酸可刺激sTNF-R的合成与分泌,而到缺血后24h,各给药组脑组织中sTNF-R的含量却表现为下降趋势。
Objective: To study the effect of the different ingredients of Qing Kai Ling Injection on the contents of TNF-α and sTNF-R in the brain of rats with MCAO,and to investigate the mec 'hanism of the different ingredients of Qing Kai Ling Injection in counteracting the damage induced by cerebral isehemic. Methods: The rats were randomly divided into two groups ( 12h and 24h of isehemia), and then divided into seven subgroups: sham-operated group, ischemic group, cholalic acid-treatment group, baicalin-treatment group, concha margaritifera usta-treatment group, jasminoidin-treatment group and the compatibility of the four ingredients -treatment group. Focal cerebral ischemia models were established by permanent occlusion of left middle cerebral artery (PMCAO) in rats. Radioimmunoassay(RIA)was used to investigate the content of tumor necrosis factor-alpha(TNF-α)in brain tissue of rats with cerebral isehemia. Enzyme-linked immunosorbent assay (ELISA) was used to detect the content of soluble tumor necrosis factor-alpha receptor. Results: After 24h of isehemia, the content of TNF-α of cholalic acid-treatment group, concha margaritifera usta-treatment group, jasminoidin-treatment group and the compatibility of the four ingredients-treatment group decreased remarkably. After 12h of isehemia, compared with model group, the content of soluble tumor necrosis factor-alpha receptor of concha morrgaritifera usta-treatment group, baicalin-treatment group and Cholalic acid-treatment group increased apparently. After 24h of ischemia, compared with model group, the content of sTNF-R in both cholalic acid-treatment group and compatible group decreased apparently. Conclusion: The four kinds of different compositions of Qing Kai Ling Injection on and the compatibility of the four ingredients can repress the synthesis and secretion of TNF-α and can inhibit the cascade reaction after cerebral isehemia. However, the effect of the different ingredients of Qing Kai Ling Injection on the content of sTNF-R is complicated. After 12h of ischemia, cholalic acid, baicalin and concha margaritifera can promote the the synthesis and secretion of sTNF-R, while the content of sTNF-R decreased in all treated group after 24h of ischemia.
