摘要
目的:对照研究自发性癫痫大鼠(SER)和正常大鼠钠通道β1亚基外显子表达情况。方法:提取鼠尾DNA,根据β1亚基外显子设计引物,进行PCR反应,琼脂糖凝胶电泳检测反应产物。PCR反应产物试剂盒纯化,送基因公司测序。结果进行Blast比对。结果:SER钠通道β1亚基2-5外显子碱基序列与原序列的同源性为100%。结论:SER钠通道β1亚基2-5外显子表达与正常大鼠无差别。
Objective: To investigate exons expression of voltage-gated sodium channel β1 subunit in spontaneously epileptic rat (SER) and Wistar rat by control study. Methods: DNA was extracted from the tail of SER. According to sodium channel β1 subunit exons design primers, sodium channel β1 subunit DNA expression was detected by PCR. PCR products were detected with gel electrophoresis. PCR products were purified and sent to Gene Company to sequencing, Results: SER sodium channel β1 subunit 2-5 exons sequence have 100% homology with the sequence of the original sequence. Conclusions: The β1 subunit 2-5 exons expression of SER has no significant difference between control rats.
