摘要
目的:在已设计合成肿瘤抗原MAGE-12的B细胞表位基础上对其进行鉴定。方法:在预测MAGE-12的B细胞表位序列为LEQRSQHCKPEE(3~14)的基础上,采用4分枝多重抗原肽结构设计合成抗原肽,免疫C-57BL/6小鼠,产生多克隆抗体,应用ELISA实验检测多克隆抗体是否为人的抗原肽的特异性抗体。结果:免疫C-57BL/6小鼠后可产生MAGE-12的抗原特异性的多克隆抗体,抗体效价最高达1:256,证明为MAGE-12抗原肽的特异性抗体。结论:证明所预测的表位LEQRSQHCKPEE(3~14)为MAGE-12的B细胞新表位,具有抗原特异性。
Objective: To synthesis and identify B-cell epitope of tumor antigen MAGE- 12, Methods: Based on epitope 3 - 14, a mutiple antigenic peptide system (MAP) was synthesized as immunogen and C-57BL/6 mice were immunised with the immunogen and the poly- clonal anti-serum was produced. The polyclonal antibody was identified by using an ELISA assay. Results: The antibody titer was reached to l: 256. The polyclonal antibedy was identified as specific antibedy of MAGE-12. Conclusion: Thoseresuhsshowedthatonepitope3- 14 may be a new B cell epitope of MAGE-12 and it is specific.
