摘要
离体大鼠工作心脏模拟体外循环缺血停搏120分钟,观察复灌早期1μmol/L腺苷的心肌保护作用。结果示实验组心功能恢复值明显高于对照组;心肌酶释放减少,冠脉流量增加;再灌45分钟,心肌ATP含量明显回升。提示复灌早期,腺苷通过改善心肌灌注,增加ATP生成等对映血再灌注心肌发挥保护作用。
The present study was undertaken to evaluate the myocardial protective effects of low-dose adenosine(AdO)added to KH buffer during the early reperfusion period.Isolated working rat hearts perfused with KH buffer,weresubjected to 120 minutes of hypothermic(15℃)cardioplegic arrest induced by St.Thomas' cardioplegic solution No2.Ingroup C(control group),the rat hearts were reperfused with KH buffer only,but in group A,AdO(1μmol/L)was added toKH buffer for the first 5 minutes of reperfusion.With another 45 minutes of reperfusion with KH buffer,recovery of func-tional indexes (as percont of preischemic control)were significantly better.The leakages of LDH and CPK were less in groupA than in group C.The coronary vascular resistance increased in hearts reperfused with ordinary perfusate but not with AdOperfusete.ATP level was decreased by ischemia and tended to be further decressed at the end of reperfusion in group C,but,was increased in group A.These data suggest that administration of AdO during the early reperfusion period might preventreperfusion-mediated myocardial injury after global ischemia and thus improve cardiac functional recovery.
