摘要
目的筛选辅酶Q10高产发酵菌种,并对发酵工艺进行优化。方法辅酶Q10发酵高产菌种采用筛选法,所产辅酶Q10类型鉴定采用硅胶薄层层析法。结果与单一碳源相比,葡萄糖、蔗糖(1∶1)的复合碳源更有利于辅酶Q10的产出;添加2%的玉米浆后辅酶Q10含量得到提高;最佳发酵pH为6.5,最佳发酵温度为30℃;对羟基苯甲酸和茄呢醇的添加都可大幅度提高辅酶Q10产量,并分别提高了11%和21%;在最优化发酵条件下,辅酶Q10产量达到188.6 mg/L。结论粉红掷胞酵母作为高产辅酶Q10菌种具有较好的实用性以及进一步提高产量的潜力和广阔的应用前景。
Aim To sieve the CoQ10 strains of high yield and optimize the fermentation conditions. Methods Sieving the coenzyme Q10 strains of high yield from the conserved strains,and CoQ10 was identified by TLC; Several factors influencing CoQ10 production were investigated. Results The results showed that the optimum fermentation conditions are as follows : carbon sources of the mixture of glucose and sucrose, nitrogen source of 2% maize liquid ; optimum pH 6. 5 and optimum temperature 30℃. The addition of precursors such as para-hydroxybenzoate and solanesol was shown to strengthen the formation of CoQ10 respectively, which were 11% and 19% higher than those under original condition; The yield of CoQ10 reached 186. 2 mg/L. Conclusion Sporobolom ycesroseus as the CoQ10 strains of high yield was practicable and marketable.
