生物免疫刺激剂对大鼠肝大颗粒淋巴细胞抑肝癌细胞增殖的影响 被引量:1

INFLUENCE OF BIOLOGICAL IMMUNE STIMULANTS ON THE CYTOSTATIC EFFECT AGAINST HUMAN HEPATOMA CELLS BY LARGE GRANULAR LYMPHOCYTES OF RAT

摘要 为从大鼠肝分离出高纯度的大颗粒淋巴细胞(LGL),以干扰素(IFN)、卡介苗(BCG)或二者联合处理LGL,用3H-TdR掺入法测定其对LGL抑人肝癌细胞增殖作用的影响。结果显示:(1)分离肝LGL电镜观察,细胞直径10~16μm,细胞表面有少量微绒毛,核多呈肾形或有切迹,胞质内可见数量不等的高电子密度颗粒。(2)LGL对人肝癌细胞有自然抑增殖作用,抑制率为27.3%,免疫刺激剂可增强LGL的抑肝癌细胞增殖作用,BCG的效应较IFN强,它们使LGL对肝癌细胞的抑制率分别提高了50%和28.6%,IFN和BCG的协同效果最强,使LGL的抑制率提高了63%。(3)电镜观察肝组织,常见LGL与枯否细胞相互毗邻。讨论了免疫刺激剂增强肝LGL和枯否细胞抗肝癌细胞作用的协同关系。 The large granular lymphocytes(Pit cells) in high purity were isolated from normal rat liver and treated in vitro respectively with IFNα 2b, BCG, or combination of both, the controls were not treated with any biological immune stimulants (BIS). The influence of BIS on LGL′s cytostatic activity against human SMMC 7721 hepatoma cells were assayed through measuring 3H TdR incorporation into tumor cell DNA. The results were as follows: (1) The ultrastructural features of isolated LGL were 10~16μm in diameter, with kidney shaped nuclei and the characteristic electron dense granules located in one side of the cytoplasm. (2) The LGL presented latent natural cytostatic activity against hepatoma cells, the inhibition rate reached 27.3%. (3)The cytostatic activity of LGL treated with BIS in vitro increased obviously, the effect of BCG was stronger than that of IFNα 2b, the effects of combined use of BCG and IFNα 2b were more effective than that of any single stimulant treatement, it increased the cytostatic activity of LGL by 63%. With the observation by transmission electromicroscope, it was found that the LGL in liver always be seen in contact with KC. Anti hepatoma effect of LGL enhanced by biological immune stimulants BIS and the relationship between LGL and KC were discussed.
