醛糖还原酶抑制剂对糖尿病大鼠肾脏超微结构的影响

EFFECTS OF ALDOSE REDUCTASE INHIBITORS ON PROTEINURIA AND GLOMERULAR ULTRASTRUCTURE IN ALLOXAN-INDUCED DIABETIC RATS

研究醛糖还原酶抑制剂防治糖尿病肾脏病变的意义。利用糖尿病ＳＤ大鼠动物模型观察了醛糖还原酶抑制剂ｓｏｒｂｉｎｉｌ对大鼠尿蛋白和肾脏超微结构的影响。结果：糖尿病鼠用ｓｏｒｂｉｎｉｌ治疗后尿蛋白呈下降趋势，肾小球基底膜厚度及系膜基质堆积明显减少。结论： ｓｏｒｂｉｎｉｌ对肾脏病变有明显疗效，使用醛糖还原酶抑制剂防治糖尿病肾病将是一条有希望的新路。

To examine the role of the polyol path way in diabetic nephropathy and evaluate the therapeutic value of aldose reductase inhibitors. METHODS Sorbinil, an aldose reductase inhibitors, was used (the daily administration, 10 mg / kg) to treat alloxan-induced SD diabetic rats over 16-week period, the animals were followed with four weekly measurement of urinary protein and blood glucose. After 16 weeks, the rats were killed and the left kidney was exised to process for electronmicroscope (EM) to determined the glomerular basement membrane width (GBMW) and area of mesangial matrix, the right kidney was placed in canoy and stained with periodic acid-schiff (PAS) for light miscroscope to determine the pathological glomerular structure. RESULTS (l) The urinary protein was partially improved in sorbinil treated groups comparing with diabetic group without treatment. (2) The peripheral GBMW and mesangial matrix were significantly lessed in sorbinil treated rats as compared with diabetic rats. CONCLUSIONS The treatment with aldose reductase inhibitors has a beneficial preventive effect on development of experimental diabetic complications.
