背景与目的:组织因子途径抑制物2(tissue factor pathway inhibitor 2,TFPI-2)是新近发现的一种丝氨酸蛋白酶抑制物,与多种肿瘤发生发展有关。本实验研究将外源性TFPI-2基因转入对胰腺癌细胞株Panc-1观察TFPI-2对该细胞细胞增殖及凋亡的影响。方法:TFPI-2基因真核表达载体、空载体分别转染Panc-1细胞,并命名为Panc-1-TFPI-2,Panc-1-V细胞与未转染的对照细胞Panc-1分别测定细胞生长曲线,DNA片段化实验检测细胞凋亡,并用流式细胞仪分析3组细胞的细胞增殖、细胞周期情况。结果:Panc-1-TFPI-2细胞同Panc-1-V和Panc-1细胞相比,细胞生长缓慢,细胞生长受到抑制,被阻滞于G0-G1期;DNA片段化实验显示Panc-1-TFPI-2细胞呈现凋亡细胞特有的“梯状”条带,而Panc-1-V和Panc-1细胞无明显“梯状”条带;流式细胞仪分析显示早期Panc-1-TFPI-2细胞凋亡率(6.9±0.5)%较Panc-1-V和Panc-1细胞凋亡率[(3.0±0.4)%,(3.5±0.4)%]增加(P〈0.05),统计学差异有显著性。结论:外源性TFPI-2基因能够抑制胰腺癌细胞生长、诱导胰腺癌细胞凋亡,可能具有抑制肿瘤细胞增殖的作用。
Background and purpose: TFPI-2 is a new serine proteinase inhibitor, it is related to many tumors. In this study, the effect of TFPI-2 gene on cell proliferation and apoptosis of human pancreatic cancer cell line Panc-1 were investigated. Methods: The growth curve was drawn for the 3 groups, Panc-1-TFPI-2, Panc-1-V and Pane-1. DNA strand breaks were used to detect the apoptosis of the 3 groups, and the change of cell cycle and apoptosis index was evaluated by flow cytometry. Results: Compared with nontransfected cells, the growth of Panc-1 cell transfected with TFPI-2 was inhibited significantly. The transfected cells showed a significant increase in G1/G0 phase. The apoptosis of Panc-1-TFPI-2 cell could be identified by DNA strand breaks and flow cytometry in comparison with the control group. The apoptosis index of the Panc-1-TFPI-2 cell (6.9±0.5) % was higher than the group Pauc-1-V and group Panc-1[ (3.0 ±0.4) % and (3.5 ± 0.4) % ] P 〈 0.05, Conclusions: The exogenous TFPI-2 gene can inhibit the growth and induce apoptosis of pancreatic cancer cell, which may also inhibit the cell proliferation of tumor cells.
