新分呼肠科病毒形态及核酸研究

THE MORPHOLOGY STUDY AND ELECTROPHORESIS ANALYSIS ON THE REOVIRUS ISOLATED FROM TICKS,BIRDS AND MICE COLLECTED IN TIBET OF OUR COUNTRY

自我国西藏边陲地区采集的蜱、鸟、鼠标本经ＢＨＫ１３细胞培养，分离到明显致细胞病变的病毒１７株。制样后电镜观察，病毒呈球形含双层壳，大小为６８～８０ｎｍ，病毒性包涵体形成明显，病毒倾向于晶格样排列，经细胞裂解而释放。病毒感染的培养悬液及蔗糖密度梯度纯化的样品均可见形态一致的病毒颗粒，其形态及形态发生符合呼肠病毒科成员的特征。纯化病毒抽提核酸，电泳结果也显相同类属的图象，说明新分病毒为呼肠病毒科成员，从其明显致病性、宿主广泛性等可能为环状病毒属，类似于科罗拉多蜱传热病毒。

Seventeen strains of virus were isolated by BHK 13 cell culture from ticks,birds and mice collected in Tibet of our country.The obvious cytopathic effect was observed in cells infected with these viruses.The viral particles were frequently found in infected cytoplasm.The viral inclusion bodies containing a number of virus particles were present throughout the cytoplasm.A lot of virus particles were tended in crystalline arrangement.The virion was released by cellular destruction.The virus particle was spherical,68～80(mean 72 4)nm in diameter with double shells.The virus particles purified in sucrose density gradient were homogeneous in shape and size.The RNA extracted from purified virion was revealed several segments with electrophoresis.The results showed that the new isolated viruses were the member of reoviridae family.The pathogenicity and replication in ticks,birds and mice make consider this virus may be similar to Colorado tick fever virus.
