摘要
为了探讨人脐血移植到小鼠体内的生长特性,将人脐血细胞注入经亚致死剂量照射后的严重联合免疫缺陷(SCID)小鼠。10只移植人脐血的小鼠中5只存活达28天,其造血和部分免疫功能得到恢复。利用人粒-单系祖细胞培养和CD+45细胞流式细胞仪检测及人HLA-DPB基因聚合酶链反应-寡聚核苷酸单链构像多态性分析等方法,检测出了植入SCID小鼠骨髓中的人脐血细胞。提示,人脐血细胞可以植入SCID小鼠并重建其造血和免疫功能。
The objective of this study was to evaluate the transplantation characteristics of human cord blood. Human cord blood cells were transplanted into sublethally irradiated severe combined immunodeficient (SCID) mice. Five out of ten transplanted mice were alived up to 28 days, both their hematopoietic and part of immunologic functions were reconstituted. With human CFU GM culture and FACS CD + 45 cells as well as HLA DPB gene PCR SSCP analysis, evidence of human cord blood cells which grew in bone marrow of mice could be detected. Hence, human cord blood cells are potential for engraftment and reconstitution of both hematopoietic and immunologic function of SCID mice. SCID mice are regarded as good model for studying the transplantation characteristics of cord blood.
