新生牛肝提取物生物学活性分析

摘要 复肝肽(FGP)系采用膜法提取技术,从新生牛肝脏中提取制备出的生物活性物质。测定其多肽的含量为950.03mg/100ml。本实验对其促生长、免疫增强作用、对化学性肝损伤辅助保护作用等生物学活性进行了研究。结果表明:小鼠体重较对照组有增加趋势(P>0.05);腹腔注射2.5、5.0、10.0ml/(kg.bw)复肝肽组小鼠玫瑰花环成环率分别高于对照组(生理盐水)33.3%,34.2%,39.6%,差异显著(P<0.05)。注射10.0ml/(kg.bw)复肝肽的小鼠血清溶血素OD值与对照组差异显著(P<0.01)。剂量组小鼠腹腔巨噬细胞吞噬百分率分别比对照组高22.1%,25.8%,26.5%,差异均有显著性(P<0.05或P<0.01),说明复肝肽具有免疫增强作用,且呈剂量依赖性。复肝肽5.0ml/(kg.bw)剂量组可有效降低四氯化碳模型小鼠血清中丙氨酸氨基转移酶(ALT),差异具有显著性(方差分析,P<0.01),病理组织学指标亦有明显改善。说明新生牛肝提取物对化学性肝损伤的辅助保护作用。 A The FGP with biological activities were extracted by membrane technology from the newborn bovine liver, with the content of polypeptides 950.03mg/100ml detected by quantitative analysis. The study were carried out on the function of the extract for growth promotion, immunological regulation,, assistant protective effect on chemical liver injury. The results showed that :The mean accumulative weight gained in polypeptide group was higher than that of the control group, but not significantly(P 〉 0. 05 ) ;The rosettes looping rates of the2.5,5.0,10.0 mL/( kg · Bw) polypeptide groups were 33.3% , 34.2% , 39. 6% higher than the control ( injected with NS) ( P 〈 0. 05 ) ;OD value of haemolysin in polypeptide group of 10ml/( kg · Bw) was significantly different from the control( P 〈 0.01 ) ;The phagocytosis percentage of the polypeptide groups were 22. 1% , 25.8% , 26.5% higher than the control group respectively ( P 〈 0. 05, P 〈 0. 01 ) ,which indicated that the polypeptide has the function of immunological regulation which presented as dose response relationship. The 5.0ml/( kg · bw) polypeptide groups can decrease the ALT in the serum of CCIA model mice obviously. The difference was serious ( ANOVA, P 〈 0. 01 ), the index of pathologic tissue became better. This show that polypeptides had assistant protective effect on chemical liver damage.
