葛根异黄酮对移植肾慢性排斥反应的防治作用 被引量:3

摘要 目的:观察葛根异黄酮对移植肾慢性排斥反应的作用,并初步探讨其机制。方法:显微外科制作Fisher-Lewis大鼠肾移植模型,受体大鼠随机分为两组,每组12只。治疗组每日灌服葛根提取物混悬液(含异黄酮5mg/mL),给药体积为1.0mL/100g体重,对照组同期给等量生理盐水,疗程为12周。分别于治疗前和治疗第4、8和12周时采尾静脉血和收集24h尿量,测血压、尿蛋白、血肌酐和血胆固醇浓度;12周时收获大鼠移植肾,行组织学和免疫组化检查。结果:与对照组比较,在治疗8周和12周时,治疗组大鼠的尾动脉收缩压、24h尿蛋白量、血清肌酐和总胆固醇浓度均明显下降(均P<0.05);治疗12周时,治疗组大鼠移植肾组织的慢性损害病理积分显著降低,肾组织的TGF-β1表达显著下调(均P<0.05)。结论:葛根异黄酮对移植肾组织的功能和结构有保护作用,能延缓慢性排斥反应的发生。 Objective To investigate the preventive effects of pueraria lobata isoflavones (PLIs) on chronic renal allograft rejection and to explore its mechanism. Methods A rat model of renal transplantation was constructed. The recipient rats were randomly assigned to receive either oral PLIs suspension at a dose of 1.0 mL/100 g (treatment group, n = 12) daily for 12 weeks, or the same volume of 0.9% saline solution (control group, n =12). Arteria caudilis systolic blood pressure (SBP), 24-hour urine protein (Upro), serum creatitine (Scr), and total cholesterol (Tch) were measured before and 4, 8, and 12 weeks after treatment. All kidney grafts were removed for histological examination and immunohistochemical detection at the end of the experiment. Results As compared with those in the control group, levels of SBP, 24-hour Upro, Scr, and Tch in the treatment group were markedly declined on weeks 8 and 12 (P 〈 0.05 for all). The chronic allograft damage index (CADI) score and TGF-β1 expressions in the renal tissues were significantly decreased in the treatment group on week 12 (P 〈 0.05 for both). Conclusions PLIs plays a protective role in the function and structure of the kidney graft. It can delay the occurrence of chronic allograft rejection.
