两种帕金森病模型黑质一氧化氮合酶mRNA的表达及灵芝孢子油的调控作用 被引量:5

摘要 目的:对比研究1-甲基-4苯基-1,2,3,6-四氢吡啶(MPTP)小鼠模型和6-羟基多巴(6-OHDA)大鼠模型黑质区诱导型一氧化氮合酶(iNOS)mRNA表达水平的变化,探讨灵芝孢子油对两种帕金森病模型黑质区iNOSmRNA表达的调节作用。方法:分别建立MPTP小鼠模型,C57BL小鼠分为MPTP组、灵芝孢子油+MPTP组、正常对照组。MPTP组皮下注射MPTP共6d,灵芝孢子油+MPTP组在注射MPTP前2d开始鼻饲灵芝孢子油,连续8d,MPTP组喂食生理盐水做对照,2周后断头取中脑腹侧组织。6-OHDA大鼠模型,SD大鼠分为6-OHDA组、灵芝孢子油+6-OHDA组、正常对照组。用脑部立体定向法将6-OHDA注射到SD大鼠一侧黑质致密部建立大鼠帕金森病模型,灵芝孢子油组在造模前3d开始给药,连续10d。6-OHDA组喂食生理盐水做对照。造模后第13天取各组大鼠中脑腹侧组织,用荧光半定量RT-PCR法检测各组中脑腹侧iNOS mRNA表达量。结果:MPTP模型中,MPTP组的iNOS mRNA表达量比正常对照组增高,灵芝孢子油组iNOS mRNA表达量比正常对照组减少,组间两两比较差异有显著性。6-OHDA模型中,6-OHDA组的iNOS mRNA表达量比正常对照组略增多,灵芝孢子油组iNOS mRNA的表达量无明显改变。两处理组组间比较无明显差异。结论:MPTP模型黑质区iNOS mRNA表达量明显增高,提示iNOS mRNA的过度表达可能与MPTP的发病机制有关,灵芝孢子油能有效下调iNOS mRNA的高表达。
