摘要
目的观察α-硫辛酸对大鼠肝脏部分热缺血再灌注损伤的保护作用。方法建立Wistar大鼠肝脏部分热缺血再灌注模型。将大鼠随机分为损伤组(A组)和处理组(B组),缺血前15 min从尾静脉分别注射生理盐水和α-硫辛酸,再灌注后1、3、6和12 h分别取血和肝脏组织,检测血清丙氨酸氨基转移酶(GPT)、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-PX),免疫组织化学检测肝脏组织中核转录因子-kB p65(NF-kB p65),RT-PCR法检测肝脏组织中巨噬细胞炎性蛋白-2 mRNA(MIP-2 mRNA)的表达。结果A组血清GPT和GSH-PX在1、3、6、12 h都明显高于B组P<0.01);A组血清GSH明显低于B组(P<0.01);B组肝脏组织中NF-kB p65和MIP-2 mRNA表达明显低于A组(P<0.01)。结论α-硫辛酸能明显减轻大鼠肝脏缺血再灌注损伤,可能与直接清除氧自由基、增加GSH产生以及减少NF-kB的活化和GSH的消耗有关。
Objective To explore the effect and mechanism of α-Lipoic acid on hepatic ischemi- a-reperfusiou injury in rats. Methods The models of liver partial ischemia-reperfusion were established in male Wistar rats. The rats were randomly divided into a control group ( group A) and an experimental group (group B) and administered either saline or α-Lipoic acid 15 min before the ischemia. GPT, GSH and GSH-PX were examined at various time points after reperfusion (1,3,6 and 12 h) in serum of the rats with or withoutα-Lipoic acid administration. The expression of NF-κB p65 and MIP-2 mRNA was detected by using immunohistochemistry and RT-PCR respectively in ischemic liver. Results Compared with group B,the ALT level and the activity of GSH-PX in serum,the expression of NF-κB p65 and MIP-2 mRNA in ischemic liver of the rats in group A were significantly increased (P 〈 0.01 ) ,while GSH in serum was decreased (P 〈0.01 ). Conclusion α-Lipoic acid may protect rats from liver ischemia-reperfusion injury through cleaning oxygen free radicals, increasing GSH production, and reducing NF-κB activation and GSH consumption.
