银杏叶提取物对幼年期大鼠惊厥后海马caspase-3FasL表达的影响 被引量:1

摘要 目的研究幼年大鼠反复惊厥后海马组织caspase-3、FasL表达的变化及银杏叶提取物干预对其的影响。方法108只20日龄健康SD大鼠随机分为三组:正常对照组、惊厥组及银杏叶提取物干预组,通过三氟乙醚反复吸入(每天1次,连续6d),制作幼年大鼠惊厥动物模型。用免疫组化(SP)法检测各组动物反复惊厥结束后1、3、7d海马组织中caspase-3和FasL蛋白的表达,RT-PCR方法检测各时相点海马组织中caspase-3mRNA的表达。结果反复惊厥结束后1、3、7d海马组织均有caspase-3、FasL的表达,与对照组比较差异有统计学意义(P<0.01)。银杏叶提取物干预组海马各时间点caspase-3mRNA及caspase-3、FasL蛋白的表达均较惊厥组显著下调(P<0.05)。结论caspase-3、FasL参与了幼年期惊厥性脑损伤。银杏叶提取物对幼年期惊厥性脑损伤的保护机制,可能与抑制海马caspase-3、FasL的异常表达有关。 Objective To investigate the expression of caspase -3 and FasL in the hippocampus of the infantile rats following recurrent seizures and the effects of Ginkgo biloba extract on them. Method 108 of 20 - day - old SD rats were randomly divided into three groups : the control group, the seizure group and the Ginkgo biloba extract treatment group. Seizures in rats were induced by inhalant flurothyl daily in six consecutive days. Brain tissue was sampled at different time points (1, 3, 7 d) after last seizure. The expressions of caspase -3 and FasL proteins in the hippocampus were detected by immunohistochemistry, and the expression of caspase - 3 mRNA was measured by reverse transcription - polymerase chain reaction (RT- PCR). Results The caspase -3 protein, FasL protein, and caspase -3 mRNA levels were im- proved obviously at post - seizure 1,3, 7 d in the hippocampus of the rat pups in seizure group. In Ginkgo biloba extract treated group, the expression of caspase- 3 mRNA, caspase- 3 protein, and FasL protein in the hippocampus at different time points significantly lower than those in seizure group (P 〈 0.05 ). Conclusions Caspase - 3 and FasL are participated in the infantile brain injury after recurrent seizures. The suppression of the abnormal levels of caspase - 3 and FasL in the hippocampus is probably related to the protective effects of Ginkgo biloba extract on the infantile brain injm'y induced by seizures.
