摘要
本试验采用鼠疫菌fra基因经PCR扩增地高辛标记方法制备基因探针,通过菌落原位杂交试验进行部分鼠疫菌、假结核菌和大肠杆菌的鉴定,被试菌中EV、EV76paris、61、M23四株鼠疫菌呈现阳性反应,假结核菌(P1,P2,P3,S-1)和大肠菌(PBR238,40,32,23)均为阴性。试验结果表明,地高辛标记的鼠疫菌fra基因探针,用菌落原位杂交试验检测鼠疫菌,其特异性强,但因本试验被试菌株数量有限,该探针能否广泛用于鼠疫防治监测,尚需进一步研究。
Y. pestis, Y. pseudotuberculosis and E.coli were identified by using the DNA probe of Y. pestis with PCR amplifications and labelled with DlG. The positive reaction occurred on EV, EV76Paris, 61, M23 of Y. Pestis and the negative reaction occurred on P1, P2, P3, S- l of Y. pseudotuberulosis and pBT238,40,32,23 of E. coli. The test indicated specificity of DNA probe fragen of Y. Pestis was high, but using the test method needs further study in plague control because the test examined the small amount bacteria.
