摘要
选用抗人T细胞CD3、CD7两株杂交瘤细胞制备合单克隆抗体(McAb)腹水,经一系列免疫化学方法纯化、鉴定后,分别以SPDP为交联剂将两种McAb与商陆毒蛋白(PAPs)偶联,构建了两种抗人T细胞免疫毒素(IT)。鉴定结果表明,IT抗体活性保持良好,并能特异而高效地杀伤T细胞及T淋巴白血病细胞CEM。本结果为探讨单抗IT联合效应及其在异体/自体骨髓净化移植治疗白血病等方面的应用研究奠定了基础。
in this study two kinds of anti-human T cell Monoclonal antibodies (McAb)-the anti-CD3 and anti-Cd7 McAbs were prepared,purified and identified as references; pokeweed antiviral protein from seeds (PAPs ) was used as ' bullets.' The anti-CD3 McAb, anti-CD7 McAb and horse anti-human thymocyte globulin (AHTG) were separately linked with PAPs by N-succinimidyl 3-(2-pyridlydithio) propionate (SPDP) and purified by Sephadex G-150. The first peaks were the immunotoxins. i. e. anti-CD3-PAPs,anti-CD3-PAPs and AHTG-PAPs. The results of SDS--PAGE analysis demonstrated that there were one or two PAPs molecules to be linked with one molecule of Ahs. The results of in vitro cytotoxicity test showed that anti-CD3-PAPs,anti-CD7-PAPs and AHTG--PAPs had a strong cytotoxic efficacy to targeted cells, i. e. T lymphocytes and CEM cells.
