摘要 目的建立HPLC法测定发酵液中苏氨酸含量的方法。方法采用OPA试剂进行柱前衍生,用氨基柱,以流动相A:PH值7.80的磷酸二氢钠溶液。流动相B:乙腈-甲醇-水为45:45:10,进行梯度洗脱,柱温40℃,检测波长:338nm,流速:2mL·min^-1,程序进样。结论浓度在0.058mg·mL^-1~1.167mg·mL^-1范围内呈良好线性(r=0.9999),重现性好,快速、简便、准确。 OBJECTIVE To set up an HPLC method to analysis the content of Threonine in fermental solution. METHODS Pre-column derivation was carried out by OPA reagent. The NH2 column was adopted, mobile phase A:sodium dihydrogen phosphate solution (pH = 7.80). mobile phase B: Acetonitrile- methanol- water = 45 : 45 : 10 column temperature was 40℃ ,detective wavelength:338nm, flow rate:2mL·mL^-1. CONCLUSION The calbration curve were linear in the range of 0. 058mg·mL^-1 -1. 167mg·mL^-1( r =0. 9999). The method was repeatable,rapid,simple and accurately.
