栉孔扇贝♀×虾夷扇贝♂单对杂交子一代的ISSR分析

ISSR analysis of the F_1 hybrids of scallop Chlamys farreri ♀×Patinopecten yessoensis ♂

用ISSR-PCR(Inter Simple Sequence Repeats PCR)技术对栉孔扇贝Chlamys farreri♀和虾夷扇贝Pa-tinopecten yessoensis♂及其单对杂交子一代担轮幼虫进行了分析。结果表明:从20个ISSR引物中筛选出4个引物对栉孔扇贝♀、虾夷扇贝♂及其单对杂交子一代担轮幼虫进行PCR扩增,共得到153个清晰稳定的扩增位点,其中母本栉孔扇贝的特异位点有38个,父本虾夷扇贝的特异位点有32个。对ISSR扩增图谱的分析结果证实,所研究的杂交子代是栉孔扇贝和虾夷扇贝的杂交种。杂交子代与母本栉孔扇贝的平均遗传相似性系数为0.6086,与父本虾夷扇贝的平均遗传相似性系数为0.4402,表明杂交子代在遗传上偏向于母本栉孔扇贝。

ISSR- PCR（ Inter Simple Sequence Repeats- Polymerase Chain Reaction）technique was used to analyse scallops Chlamys farreri ♀ , Patinopecten yessoensis ♂ and their hybrids in order to probe into the application of this technique in scallop hybrid identification and verifying the sibship between parents and hybrids. Four primers screened out from twenty ISSR primers were employed to amplify the genome DNA of Chlamysfarreri ♀ , Patinopecten yessoensis ♂ and their hybrids. 153 distinct and highly reproducible ISSR bands were detected, in which 38 bands were the special bands of Chlamys farreri ♀ and 32 bands were the special bands of Patinopecten yessoensis ♂. The ISSR electrophoretogram confirmed that the filial generation was the hybrids of Chlamys farreri ♀ and Patinopecten yessoensis ♂. The average genetic similarity coefficient between hybrids and Chlamys farreri ♀ was 0. 6086, and the average genetic similarity coefficient between hybrids and Patinopecten yessoensis ♂ was 0. 4402, indicating that the hybrids were genetically closer to their maternal stock Chlamys farreri ♀.
