健脾补肾、和解泄浊法抑制肾间质纤维化的实验研究 被引量:4

Effects of Reinforcing Kidney and Exhausting Toxic Mixture on Renal Interstitial Fibrosis in Rats

摘要 目的观察补肾排毒合剂对单侧输尿管梗阻(UUO)后大鼠肾间质α-平滑肌肌动蛋白(α-SMA)、Ⅳ型胶原(ColⅣ)表达的影响及对肾间质纤维化的保护作用。方法采用UUO诱导肾间质纤维化的动物模型,将48只Wistar大鼠随机分为假手术组(Sham)、模型组(UUO)和补肾排毒合剂治疗组(REM),每组分别于造模后3、10、20、30d各处死4只,取左肾组织行HE染色,动态观察肾脏病理学变化,评定各组肾小管间质损害程度,以免疫组化方法测定肾组织中α-SMA、ColⅣ。结果与UUO组比较,REM组大鼠肾间质病变程度明显减轻,α-SMA、ColⅣ在肾间质中的表达也有明显降低。结论补肾排毒合剂可通过降低α-SMA、ColⅣ的表达,减少肾间质细胞外基质合成,延缓肾间质纤维化的进展。 Obujective: To explore the preventive and theraputic effects of Reinforcing Kidney and Exhausting Toxic Mixture(REM) on renal interstitial fibrosis by unilateral ureteral obstruction. Methods: Rar unilateral ureteral ohstruction(UUO) model was used for the study. 48 Wistar rats were randomly divided into 3 Groups of 16 rats: Shan - oparation, UUO and UUO + REM. For rats from each groups were sacrificed at 3, 10, 20, 30days after U- UO. left kidney of each rats was stained with hematoxylind eosin method. Evaluate the degree of tubula - interstitial damage. α- smooth muscle action(α- SMA) and collagen IV were identified using an imunohistochemistry tech- nique. Reults: The real pathologic lesions were significantly alleviated in REM treated rats as compared with those inflicted with UUO. Immunohistochemisry staining studies also showed a marked decrease in the expression of α - SMA and collagen IV in. Conclusions: REM cold renard the expression of α - SMA and collagen IV in renaltissues, which may decrease matrix secretion and retard progression of renal interstitial fibrosis.
