真核表达质粒pSG.SS.C3d3.YL.F_(β.ECD)免疫避孕效应研究

摘要 目的研究鼠受精素β胞外区(Fβextracellular domain,F_(β.ECD))真核表达重组质粒pSG.SS.C3d3.YL.F_(β.ECD)的DNA疫苗免疫避孕效应。方法应用pSG.SS.C3d3.YL.F_(β.ECD)(C3d3-YL-Fβ组)肌注雌鼠,观察其免疫避孕效应,ELISA法检测雌鼠血清抗Fβ蛋白IgG抗体最大稀释度、IL-2、INF-γ、IL-4、IL-10水平和阴道分泌物抗Fβ蛋白IgA抗体最大稀释度。对照组为PBS组、空载体pSG.SS.YL组(空载体组)和pSG.SS.YL.Fβ.ECD(YL-Fβ组)。结果C3d3-YL-Fβ组雌鼠生育能力显著降低,雌鼠妊娠率显著降低,一胎产仔数量显著降低,免疫避孕效果显著好于YL-Fβ组。pSG.SS.C3d3.YL.F_(β.ECD)DNA疫苗可激发雌鼠产生高水平血清抗Fβ蛋白IgG抗体,阴道分泌物中出现抗Fβ蛋白IgA抗体,血清IL-2、INF-γ浓度较对照组显著降低,血清IL-4、IL-10浓度显著增加。结论真核表达重组质粒pSG.SS.C3d3.YL.Fβ.ECD可有效诱导雌鼠免疫性避孕效应,效果优于pSG.SS.YL.F_(β.ECD)。 Objective To explore the immunological contraceptive effects induced by pSG.SS.C3d3.YL.F β.ECO expressing mouse sperm fertilin β subunit protein. Methods The female mouses were immunized with pSG.SS. C3d3.YL.F β.ECD. Immunocontraceptive effects of pSG.SS.C3d3.YL.F β.ECD were detected. Their serum IgG titres and vaginal secretions IgA titres against to F β protein were detected using ELISA assays. The content of IL-2, INF-γ, IL- 4 and IL-10 were detected using ELISA assays too. The control groups were PBS group, pSG.SS.YL group and pSG. SS.YL.F β.ECD group (YL-F β group). Results The pregnant rate of the mice immunized by C3d3-YL-F β group was significantly lower than that in the YL-F β group. The fertility of the two immunized groups was significantly decreased than those in the PBS group and YL group. Higher level serum IgG titres in mice induced by DNA immunization were detected in C3d3-YL-F β group. In the C3d3-YL-Fβ group, the content of IL-2 and INF-γ decreased significantly. The content of IL-4 and IL-10 increased significantly. Conclusion pSG.SS.C3d3.YL.F β .ECD can effectively induce more specific humoral and mucosal antibody response and contraceptive effects in Kunming mice than pSG.SS.YL.F β .ECD.
