盱眙水牛病病原研究初报

摘要 采取人工传代,部分原代细胞或传代细胞系分离培养、超薄切片电镜观察以及免疫沉淀反应等方法对盱眙水牛病的病原进行研究。结果表明,将患有该病的水牛经过全血静脉接种在健康牛体内,可使其人工感染发病并长期继代。但随继代次数增高,病水牛病程延长,且部分病变不明显。所用的细胞培养均未导致细胞病变,亦未从细胞培养中检出抗原。10例自然发病和人工发病水牛中,有4例从淋巴结、5例从白细胞的电镜超薄切片中发现了大小约100nm、有囊膜的圆形病毒颗粒,并有病毒成熟出芽的图像。病水牛的淋巴结、白细胞等组织提取物中亦存在上述病毒颗粒。其提取物具有特异性抗原,可以与本病后期水牛血清发生特异性免疫沉淀反应。初步认为该病毒是盱眙水牛病的病原体,属一种慢病毒。 Xuyi Buffalo Disease is an acute or subacute febrile infection limited in buffalo, it was first found in Xuyi county, Jiangsu province, China. The article reported that with artificial inoculation of the whole blood from sicked buffalo experimentally, the susceptible animals could appear the same symptom. The whole blood collected from one sicked buffalo had been passed over 35 times and 97/100 of the susceptible animals were sicked. The inoculation period ranged from 10 to 120 days, and 46 days on the average, but as the time continued, the sicked appeared few changes and the sicked period was longer,the mortality of the disease kept almost 100 percent. EM observation of the lymph nodes, white cells or liver tissues of 10 samples, the virus was found to be two figures of type C and mature type B with about 100 nm in diameter and outer membranes in 5 cases from white cells and 8 cases of lymph nodes, livers and spleens, also found that the budding process was in the same cases. By the cell culture of different cell systems such as MA104,BHK,PK 15, Vero or the primary kidney cells of rabbits and balf, CPE or virus particles were not found until now. Using immunization method, the antigen that from the suspension of the lymph node, liver and spleed were centifugen with different rate could react with the later sera of the sicked and affected ones (10/12)specially. Two positive sera also reacted with the tissue suspension of 7 sicked. We considered that the virus may be the agent of Xuyi Buffalo Disease and it could belong to one kind of the retrovirus.
