EMMPRIN、MMP-2和MMP-9在恶性淋巴瘤中的表达及其临床意义 被引量:1

摘要 目的探讨恶性淋巴瘤中细胞外基质金属蛋白酶诱导因子(EMMPRIN)的表达及其与基质金属蛋白酶(MMPs)表达和疾病进展的关系。方法运用实时定量PCR法检测了81例恶性淋巴瘤患者淋巴瘤组织中EMMPRIN、MMP-2和MMP-9的表达。同时通过电镜对淋巴瘤组织切片中肿瘤浸润血管情况进行观察。结果与淋巴结反应性增生(8例)相比,不同类型的恶性淋巴瘤均高表达EMMPRIN、MMP-2和MMP-9。EMMPRIN来自淋巴瘤细胞。Ann Arbor分期为III-IV、伴多个淋巴结外病变和国际预后指数分组为高危组的患者上述基因的表达水平显著升高。此外,EMM-PRIN、MMP-2和MMP-9高表达者可见淋巴瘤细胞浸润血管。结论EMMPRIN表达可促进恶性淋巴瘤MMPs的表达,并与疾病进展密切相关。 Objective The study was aimed to investigate the extracellular matrixmetalloproteinase inducer expression (EMM- PRIN) in lymphoma, as well as its clinical significance and relation to matrix metalloproteinases (MMPs) expression. Methods EMMPRIN, MMP-2 and MMP-9 mRNA expression from lymphoma tissue of 81 patients were measured by real-time quantitative PCR. Meanwhile, lymphoma infiltration on tumor vessels was identified by transmission electron microscopy. Results Comparing with 8 patients with reactive hyperplasia,EMMPRIN,MMP-2 AND MMP-9 were overexpressed in different histological types of lyrnphoma. EMM- PRIN was derived from lymphoma cells. Increased EMMPRIN, MMP-2 and MMP-9 levels were associated with advanced Ann Arbor stage, multiple extranodular involvement and high-risk international prognostic index. In cases overexpressing EMMPRIN,MMP-2 and MMP-9,ultrastructural study showed vessels invaded by lymphoma cells. Conclusion The value of EMMPRIN expression as a biomarker of disease progression in lymphoma should be considered.
