摘要 目的:观察体外雪旺细胞环境对大鼠骨髓间充质干细胞(MSCs)的诱导分化作用。方法:分离和体外培养新生大鼠MSCs和雪旺细胞;实验组MSCs经DAPI标记后与雪旺细胞按1∶2比例共培养,观察细胞的形态变化,在第1,3,6天时使用免疫细胞化学染色检测MSCs中Nestin,GFAP及S-100表达。对照组MSCs单独培养,其余处理与实验组相同。结果:共培养1 d后部分MSCs出现雪旺细胞样形态,Nestin表达率为(83.4±3.5)%,随时间增加而降低。GFAP及S-100表达率随时间而增加。对照组细胞形态无变化,不同时间点均有少量MSCs表达Nestin,而GFAP和S-100未见表达。结论:MSCs可通过体外共培养向雪旺细胞分化。 Objective: To investigate the inducing effect of Schwann cells environment on the differentiation of bone marrow mesenchymal stem cells (MSCs) in vitro. Methods: MSCs and Schwann cells were isolated from newly-born rats and cultured in vitro. In treatment group, MSCs were labeled by DAPI and co-cuhured with Schwann cells (1 : 2 ratio). In control group, MSCs were solely cultured. The morphological change of the cells was observed. The expression of Nestin, GFAP and S-100 was detected by immunocytochemical staining at 1, 3 and 6 d after co-cuhure. Results: The MSCs changed morphologically into Schwann-like cells in co-cuhure group at 1 d, the positive rate of Nestin was(83.4±3.5)%, then decreased. The expressions of GFAP and S- 100 were increased with time. In control group, no morphological change was observed, Nestin expressed at very low level while GFAP and S-100 almost were not expressed. Conclusion: MSCs can be induced to differentiate into Schwann cells by co-cuhure in vitro.
