摘要 目的:研究PEG10基因在肝细胞肝癌(HCC)中的表达及其与凋亡的相互关系,探讨其在HCC发生发展中的作用及意义。方法:应用半定量逆转录-聚合酶链反应(RT-PCR)法定性测定43例HCC及对应癌旁组织中PEG10 mRNA的表达情况,TUNEL法原位检测HCC组织中的凋亡癌细胞并计算凋亡指数(AI)。结果:HCC细胞中PEG10 mRNA的阳性表达率明显高于对应的癌旁组织(P<0.01);PEG10 mRNA的表达水平与患者的年龄、性别、肿瘤大小无关(均为P>0.05);但与肿瘤的分期分级、分化程度、浸润转移、门静脉有无癌栓、血清中甲胎蛋白水平、是否合并HBV感染有关(均为P<0.05);AI在PEG10表达阳性与阴性组间差异有显著性(P<0.01)。结论:PEG10在HCC中过度表达,抑制肝癌细胞凋亡,可能在HCC的发生发展中起着重要作用。 Objective: To investigate the relationship between expression of PEG10 gene and apoptosis in human hepatocellualr carcinoma (HCC) and discuss its significance in the hepatocellular carcinogenesis. Methods: Expression of PEG10 mRNA in HCC and the paired para-cacinoma liver tissues from 43 HCC patients was detected by reverse transcription polymerase chain reaction (RT- PCR). Apoptosis index was detected by TUNEL method. Results: The positive rates of PEG10 mRNA in HCC were significantly higher than those in the paired para-carcinoma liver tissues(P〈0.01). Chi-square test analysis revealed that increased PEG10 mRNA expression was correlated with tumor clinnical grade and stage, differentiation, invasion and metastasis, portal cancer embolus, positive HBsAg and high AFP level(all P〈0. 05). The average index of apoptosis in PEG10 mRNA positive group was significantly lower than those in negative group(all P〈0.01). Conclusion: The over-expression of PEG10 can inhibit cell apoptosis in HCC, and the results indicate that PEG10 participates in occurrence and development of HCC.
