摘要
目的:研究丁酸钠对正常和炎症结肠上皮细胞凋亡的影响及作用机制。方法:离体大鼠结肠黏膜固定于Ussing槽中孵育,黏膜面给予4%乙酸,并与正常结肠黏膜对照;在体实验,大鼠用醋酸诱导大鼠结肠炎。3 d后随机分为两组,每天分别给予丁酸钠、生理盐水灌肠1次。于第11天处死,取结肠组织进行TUNEL检测及免疫组化分析结肠上皮细胞凋亡情况。结果:①乙酸孵育无丁酸钠组离体结肠上皮细胞凋亡指数为25.37±2.38,乙酸孵育加丁酸钠组凋亡指数为10.58±1.77,P<0.05。正常黏膜无丁酸钠组结肠上皮凋亡指数为10.35±1.07,正常黏膜组加丁酸钠组结肠上皮凋亡指数为3.78±0.70,P<0.05。②丁酸钠灌肠治疗组平均凋亡指数2.35±0.41,凋亡指数和Bax表达水平明显低于对照组18.9±1.72,P<0.01。结论:似于体内肠腔正常浓度的丁酸钠(10 mmol/L)在体外体内对正常和炎症结肠上皮细胞有抑制其凋亡的作用。
Objective: To observe the effect of sodium butyrate in the apoptosis of normal and inflammate colonic epithelial cell. Methods: (1) The rat's colonic mucosa was suspended in Ussing Chamber. The mucosa side was treated with 4% acetic acid + Ringer solution and the rat's self blood, (2) Distal colitis in vivo was induced in rats with an acetic acid enema for three days, and the rats were randomly divided into two groups according to the treatment of 40 mmol/L Na-butyrate enemas and saline enemas daily. Colonic tissues were examined by immunohistochemistry at 11 d. Results. (1) After incubated with acetic acid and without sodium butyrate, the apoptosis rate of colonic epithelium cells was 25. 37±2.38, which was higher than that treated with sodium butyrate(10.35±1.07, P〈0.05). The apoptosis rate of normal mucosa treated without sodium butyrate was 10. 35±1.07 and was higher than that treated with sodium butyrate (3. 78±1.07, P〈0.05). (2) The apoptosis index and the expression of Bax was significantly decreased in the butyrate enemas group comparing with those in the saline enemas groups. Conclusion: Sodium butyrate could inhibit the apoptosis of colonic epithelium cell in vitro and in vivo.
