摘要
目的在原位建立膀胱肿瘤的大鼠模型。方法实验动物随机分为两组,模型组55只,对照组10只,N-甲基亚硝基脲(MNU)对模型组SD大鼠进行膀胱灌注,于首次灌注MNU后第3、6、9周分别每次处死模型组10只大鼠以及对照组2只大鼠,在病变明显处多处取材进行组织病理学检查。在实验的12周处死剩余的模型组和对照组大鼠进行组织病理学检查。结果诱癌期间两组大鼠毛色、食量、营养状况及体重等一般情况无明显差别;对照组大鼠膀胱黏膜未见病变,12周时,所有模型组大鼠均有肿瘤形成且大多呈多发性,肿瘤大小不一;对照组大鼠膀胱黏膜上皮细胞排列有序,形态大小未见异常。模型组均发生程度不等病变,12周时肿瘤细胞浸润可达深肌层,肿瘤细胞异型明显,细胞核较大且极性紊乱或消失,核分裂相明显。结论成功在原位建立膀胱肿瘤的大鼠模型。
Objective To construct the rats bladder turner model in situ. Methods Experimental animals were divided randomly into two groups,55 rats in model group and 10 rats in control group. Model SD rats were irrigated MNU in bladder,model group 10 rats and control group 2 rats were executed respectly every time in 3 , 6, 9 week after irrigating MNU first time. The histopathology checking was carried out on poly-passim obviously pathological changes passim. Surplus rats of model group and control group were executed in experimental 12 week and carried out histopathology checking. Results General states of health of two groups rats, concluding color pattern, appetite, nutritional state and body weight, were not significantly different during luring cancer. In 12 week,all model group rats had turner formation and most turners assumed multiple and size disparation. Bladder mucosa epithelial cells of control rats assumed order arrangement and no abnormality seen in shape and size. All model group rats developed disparat degrees pathological changes , turner cells infiltrated into deep muscularis in 12 week, turner cells had obviously allotype,cell nucleus were more bigger and polarity chaos or disappearance,caryocinesia phase manifest. Conclusion Rats were con- structed succeedly bladder turner model in situ.
