Heparanase与bFGF在膀胱移行细胞癌中的表达研究

摘要 目的探讨类肝素酶(heparanase,Hpa)与碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)在膀胱移行细胞癌(transitional cell carcinoma of the bladder,TCCB)发生、发展中的作用及意义。方法应用免疫组织化学S-P法,检测69例BTCC和9例正常膀胱组织中Hpa和bFGF的表达。结果膀胱癌中,Hpa阳性表达率为42.03%;bFGF阳性表达率为44.93%;Hpa与bFGF共表达阳性率为31.89%;9例正常膀胱黏膜中均未见Hpa和bFGF阳性表达。Hpa与bFGF表达均随着膀胱癌病理分级和临床分期的升高而增强(P<0.05)。结论Hpa与bFGF可能作为预测膀胱移行细胞癌进展的指标及肿瘤治疗靶点,并有可能成为一个有效的预后指标。 Objective To assess the expression of heparanase and basic fibroblast growth factor(bFGF) in transitional cell carci- noma of the bladder(TCCB) and their relation to tumor occurrence and development. Methods A total of 69 cases of TCCB and 9 normal bladde paraffin-embedded tissue specimens were studied by immunohistochemistry streptavidin-biotin peroxidase(SP) method to evaluate the expression of heparanase and bFGF. Results Expression of heparanase and bFGF was detected in 42.03% and BTCCs in 44.93 % respectively, while none expression in normal bladder epithelium. Co-expression rate of heparanase and bFGF was 31. 89%. Furthermore, the expression of heparanase and bFGF was significantly higher in higher clinical stage and pathological grade tumors(P〈0.05). Conclusion Up-regulation of heparanase and bFGF expression indicates that they may be involved in tumor cell differentiation,invasion and metastasis. Heparanase and bFGF may serve as predictors of progession in TCCB and good target for tumor therapy, and they also seem to be effective prognosis markers of TCCB.
