摘要
目的:探讨TGF-β及其伙伴分子Smad4在胃癌组织中的表达。方法:利用逆转录(RT)-PCR技术测定胃癌组织TGF-β mRNA和Smad4 mRNA水平,并分析两者与胃癌患者各项临床病理指标间的关系。结果:Smad4 mRNA在胃癌组织中的阳性表达率明显下降。TGF-β和Smad4均与胃癌组织的分化程度、肿瘤大小、浸润深度、淋巴转移和TNM分期有关。Smad4 mRNA在胃癌组织分化程度高、肿瘤<5cm、浸润深度浅、无淋巴转移、TNM分期较早的患者中阳性表达率高。TGF-β mRNA在胃癌组织中的阳性表达率明显升高。TGF-β mRNA在胃癌组织分化程度高、肿瘤<5cm、浸润深度浅、无淋巴转移、TNM分期较早的患者中阳性表达率低。胃癌组织中TGF-β和Smad4表达间呈负相关。结论:TGF-β和Smad4共同调节胃癌的发生、发展及其生物学行为。
Objectives: To explore the correlation of Smad4 mRNA and TGF-β mRNA in gas-tric carcinoma. Methods: The presence of Smad4 mRNA and TGF-β mRNA was measured by re-verse transcriptase polymerase chain reaction (RT-PCR) from 61 gastric carcinomas and 39 normal controls. Results: The positive percentages of Smad4 mRNA in gastric carcinoma were lower than those of normal controls. The positive percentages of TGF-β mRNA in gastric carcinoma were high-er than those of normal controls. The expressions of Smad4 mRNA and TGF-β mRNA were all re-lated to histological differentiation, location, tumor size, infiltration, lymph node metastasis and TNM stage. Negative correlation occurred in Smad4 mRNA and TGF-β mRNA. Conclusion: TGF-β and Smad4 may be associated with the carcinogenesis, biological behaviors and prognosis of gastric carcinoma.
