摘要
目的:建立油酸诱导的慢性胰腺炎大鼠继发糖尿病模型,为探讨胰源性糖尿病的可能机制、药物防治胰源性糖尿病的研究奠定基础。方法:选用健康雄性Wistar大鼠经肠壁逆行胰胆管插管,经插管向胰腺内逆行灌注油酸的方法制作慢性胰腺炎大鼠继发胰源性糖尿病模型。对照大鼠经插管灌注等量的生理盐水。6周后观察大鼠体重、胰腺形态、病理及超微结构学改变、糖耐量、空腹血糖、淀粉酶、胰岛素和胰高糖素变化。结果:模型组较对照组的体重减低,胰腺形态外观糜烂,病理及超微结构退行性改变,外分泌腺体广泛退化,内分泌腺体继发损伤,淀粉酶浓度降低,空腹血糖升高,糖耐量异常,胰岛素降低,胰高糖素降低。结论:油酸诱导的慢性胰腺炎大鼠继发胰源性糖尿病模型适合进行胰源性糖尿病的研究。
Objective: To establish model of pancreatogenic diabetes developed from chronic pancreatitis induced by oleic acid on rat, discuss mechanism of overt pancreatogenic diabetes, and provide foundation for the medical research of pancreatogenic diabetes. Methods: An experimental model of pancreatogenic diabetes developed from chronic pancreatitis was induced by oleic acid retrogradely injected into the pancreatic duct of rat. Pathological and uhrastructural changes, amylase, glucose tolerance test, glycemia, circulating insulin and glucagons were measured 6 weeks later. Results: The rats of chronic pancreatitis induced by oleic acid demonstrated remarkable weight loss, appearance, ultrastructure, pathological of pancreas, extensive acinar degeneration, overt damage of islet and remarkable changes of amylase, glucose tolerance, glycemia , insulin, and glucagons. Conclusion: The model of pancreatogenic diabetes developed from chronic pancreatitis induced by oleic acid on rat is suitable for research of pancreatogenic diabetes.
