摘要
目的选择在肿瘤血管形成过程中较为重要的因子即血管内皮生长因子(vascularen-dothelialgrowthfactor,VEGF)为靶,进行抗肿瘤形成的实验性研究。方法应用亲和层析纯化的有中和活性的VEGF抗体进行实验性肿瘤的抑制研究。结果抗VEGF抗体对小鼠S180肉瘤的抑制作用呈明显的剂量依赖关系,当剂量为每只200μg/d时,抑瘤率为41.0%,在裸鼠移植人胃癌(MGC803)模型中,VGEF抗体的抑瘤率可达76.2%,如合并使用胃癌单克隆抗体与131I偶合物(131I-3H11),可使4/5的裸鼠无瘤生存,其抑瘤率较单独使用偶合物增加84.0%。结论VEGF抗体对肿瘤治疗具有潜在的应用价值。
Objective Vascular endothelial growth factor(VEGF) is a very important factor in the process of angiogenesis,which is chosen as a target for experimental study of anticancer. Methods Anti VEGF antibody was obtained from immunized rabbits and purified with affinity col umn.Morever,it was identified as neutralized antibody by Mile′sassay.Results he anti VEGF antibody was able to inhibit growth of S180sarcoma in adose dependent manner,and the inhibition rate reached41.0%with dose200μg per mouse per day.For human gastric cancer(MGC 803) bearing nude mice,Anti VEGF antibody inhibited tumor growth by76.2%.When anti VEGF anti body was combined with 131 I 3H11,amurine monclonal antibody conjugated with 131 Ifor treatment, only one of five nude mice developed tumor and84.0%more inhibition of tumor growth was obtained in comparison with treatment by 131 I 3H11only.Conclusion The anti VEGF antibody may be poten tially useful for clinical treatment of cancer.
