摘要
用15只大鼠作在体心脏缺血再灌模型,研究了心肌间质缺血再灌损伤和维拉帕米抗缺血再灌损伤的作用.实验分3组:A组(缺血再灌组),B组(维拉帕米组)和C组(假手术组).电镜下见A组胶原纤维明显减少,毛细血管基底膜缺损,内皮细胞吞饮小泡减少,间隙增宽;B组胶原纤维减少和毛细血管损伤不明显.超微结构立体计量显示,A组胶原纤维线密度降低(P<0.05).羟脯氨酸测定发现,A组含量减少(P<0.05).结果表明:心肌缺血再灌可致间质损伤,维拉帕米能减轻间质缺血再灌损伤.
The myocardial interstitium ischemia-reperfusion injury and protective effects of Verapamil against ischemia-reperfusion injury were studied in vivo ischemia-reperfusion model of rat heart. Fifteen male SD rats were randomly divided into three groups, ischemia(60 min)-reperfusion(120 min) group (Group A), Verapamil-treated group (Group B) and sham-operation group ( Group C). Electron microscopic observation showed that the collagenous fibers decreased in Grou A. Structural changes in capillaries of Group A included loss of endothelial pinocytotic vesicles, widening of gaps within the endothelium, and loss of lamina. As compared to Group A, the decrease of collagenous fibers and the damage of capillaries in Group B were not evident. Ultra-structure stereological quantitative assay revealed that the length density of collagenous fibers and the content of hydroxyproline decreased significantly in Group A (P<0. 05). The above results suggested that myocardial interstitium was damaged by ischemia-reperfusion and that Verapamil could protect the myocardial interstitium against ischemia-reperfusion injury.
