摘要
目的:研究重组人巨噬细胞集落刺激因子(rhM-CSF)对骨髓造血功能恢复的影响。方法:以亚致死剂量60Coγ射线照射BALB/c小鼠,造成放射性损伤,继以家蚕表达的rhM-CSF连续7天给每只小鼠腹腔注射进行治疗。结果:照射后第15和20天,治疗组小鼠外周血白细胞计数(×109/L)为3.42±1.20和5.56±2.50,显著高于对照组的2.03±0.90和3.72±2.30(P均<0.05);照射后第10,15,20,25和30天,治疗组小鼠外周血单核细胞计数(×109/L)为0.08±0.06,0.16±0.10,0.48±0.35,0.47±0.21和0.33±0.17,分别高于对照组的0.025±0.016,0.05±0.04,0.23±0.16,0.33±0.19和0.17±0.13(P均<0.05);照射后第15,20和25天,治疗组小鼠外周血中性粒细胞数(×109/L)为1.03±0.61,2.18±1.19和3.28±1.09,也高于对照组的0.62±0.37,1.40±0.99和2.20±0.74(P均<0.05);照射后第9天骨髓CFU-GM集落形成试验显示对照组的集落数(19±11/?
Objective:To demonstrate the hematopoietic enhancement activity of recombinant human macrophage colony stimulating factor (rhM CSF). Methods:Balb/c mice were sublethally irradiated with 60 Co γ rays and then the rhM CSF expressed in silkworm (10 3U per individual for 7 days) was administered intraperitoneally.Results:①The peripheral white blood cell counts of treatment group at day 15 and day 20 after irradiation were significantly higher than those of normal control group;②At days 10,15,20,25 and 30 after irradiation,the difference between the peripheral monocyte counts of the treatment and control group were statistically significant;③At days 15,20 and 25 after irradiation, the difference between the peripheral granulocyte counts of the two groups were also significant;④At day 9 after irradiation,the bone marrow CFU GM yield of the control group were significantly lower than those of the treatment group.Conclusion:rhM CSF expressed in silkworm could accelerate hematopoietic recovery in irradiated mice.
