正常豚鼠耳蜗Corti器CaM-PK Ⅱ的分布及活性

摘要 目的探讨CaM-PKⅡ在豚鼠耳蜗基底膜中的分布、活性及其意义。方法通过免疫组化及放射免疫、扫描电镜进行研究。结果正常豚鼠耳蜗Corti器各回内、外毛细胞内CaM-PKⅡ呈中等强度免疫阳性反应,OHC分布较IHC稍高。其活性以Ca2+/CaM依赖为主,占94.74%。结论CaM-PKⅡ与耳蜗结构紧密相关。 Objective To evaluate dynamic changes of CaM-PKⅡ in cochlear of guinea pigs. Methods The expressions of CaM- PKⅡ in cochlear basilar membrane were assessed by immunohistochemistry,32P-ATP incorporation and scanning electron microscope. Results By immunohistochemistry staining,CaM-PKⅡ positive reaction was found in HC of normal guinea pigs,with more intensively positive reactions in outer hair cells ( OHC ) than that in inner hair cells ( IHC ), which widespread in cytoplasm and negative in nucleus. The total activity of CaM- PKⅡ in cochlear basilar membrane of guinea pig was ( 3. 255 ± 0.036 ) pmol·min^-1·mg-1, which includes Ca^2+/CaM dependent activity (94.74%) and Ca^2+/CaM independent activity (5.26%). Conclusion Changes of CaM-PK Ⅱ may be associated with the cochlear.
