寡聚脱氧核苷酸对BALB/c小鼠Th细胞的调节

摘要 目的观察在体外实验中,包含有重复序列TTAGGG的寡核苷酸(ODN_(A151))对小鼠脾淋巴细胞分泌Th1、Th2型细胞因子的影响。方法分离小鼠睥淋巴细胞,根据分组加入伴刀豆蛋白A(ConA)以及不同浓度的ODN_(A151)或对照的ODN_(1612),培养72 h后采用夹心法ELISA测定上清中的IFN_γ和IL-4浓度。结果ODN_(A151)抑制淋巴细胞培养上清的IFNγ浓度,并升高IL-4的浓度,降低IFN_γ与IL-4的比值,并且ODN_(A151)的作用呈剂量依赖关系;而对照的ODN_(1612)对细胞因子水平无影响。结论在非抗原特异性、多克隆刺激剂(ConA)作用下,ODN_(A151)对小鼠睥淋巴细胞的Th1/Th2平衡有调节作用,既抑制Th1分化又促进Th2分化。 Objective To explore the effect of oligodeoxynucleotides (TTAGGG) on the cytokines secreted by splenetic lymphocytes of BALB/c mice. Methods Firstly, separated splenetic lymphocytes of BALB/c mice; after cultivation of 72 hours with concanavalin A (ConA) and several concentration of (1 μM, 5 μM, 10 μM)ODNA151 or ODN1612(10 μM), then detected IFN γ/and IL-4 in supernatant by ELISA. Results ODNA151 but not ODN1612 significantly inhibited IFNγ/and increased IL-4 (P 〈 0.01 ), accordingly down-regulated ratios of IFNT/IL-4; This effect was linear correlated on the concentration of ODNA151. Conclusion On stimulation of ConA, ODNA151 regulated differentiation of Th lymphocytes and skewing Th1/ Th2 balance toward Th2 type eytokine production.
