活血潜阳颗粒对自发性高血压大鼠血浆肾素血管紧张素系统的影响 被引量:13

Effects of "Huoxue Qianyang Granule" on Renin-Angiotensin System in Plasma of Spontaneous Hypertensive Rats

摘要 目的观察活血潜阳颗粒对高血压大鼠血浆肾素血管紧张素系统的影响,探讨其作用机制。方法以自发性高血压大鼠(SHR)为高血压及高血压左心室肥厚模型,随机分为活血潜阳颗粒大、中、小剂量组,卡托普利组,松龄血脉康组,双蒸水组;并以Wistar-kyoto(WKY)大鼠为正常对照组。9周后检测大鼠血浆肾素(PRA)、血管紧张素转换酶(ACE)活性以及血管紧张素Ⅱ(AngⅡ)含量。结果与WKY大鼠相比,SHR双蒸水组ACE显著升高;活血潜阳大剂量组明显降低SHR血浆ACE活性,降低值低于卡托普利组,两组之间差异无显著性。与松龄血脉康组相比,活血潜阳颗粒对ACE活性影响更大,但差异无显著性。与WKY大鼠相比,SHR双蒸水组血浆肾素(PRA)活性升高,但差异无显著性;各药物对PRA活性影响不明显。与WKY对照组比较,SHR双蒸水组血浆AngⅡ含量升高,活血潜阳中剂量组能明显降低SHR血浆AngⅡ的含量(P<0.05),其降低值不及卡托普利组,但两者组间比较无显著性差异(P>0.05);与松龄血脉康组相比,活血潜阳颗粒对AngⅡ降低作用更大,但差异无显著性(P>0.05)。结论活血潜阳颗粒可抑制高血压大鼠血浆ACE活性,减少AngⅡ的生成,可能为该方控制血压、逆转左心室肥厚作用的另一机制。 Objective To study the effects of "Huoxue Qianyang Granule" (HXQY) on renin-angiotensin system in spontaneous hypertensive rats (SHR) to investigate its mechanisms. Methods Rats of spontaneous hypertension and LVH were randomized into 7 groups: distilled water, "Songling Xuemai Kang"(SLXMK), Captopril, large or middle or little dosage HXQY and WKY rats were used as normal group. ACE activity, PRA, and Ang Ⅱ levels in plasma were measured after 9-week treatment. Results Plasma ACE activity, PRA, and Ang Ⅱ levels in SHR were increased; HXQY could descend ACE activity and Ang Ⅱ levels remarkably, but no influence on PRA; ACE activity of large dosage HXQY was lower than SLXMK, Ang Ⅱ levels of middle dosage HXQY was lower than SLXMK, but no marked difference between HXQY and SLXMK; all of them were higher than Captopril, but no remarkable difference. Conclusion Restricting the ACE activity and decreasing Ang Ⅱ levels in plasma is another mechanism of HXQY in controlling the blood pressure and reversing left ventricular hypertrophy.
