摘要
以盐生动物卤虫为材料,分别用氯化钠高盐沉淀法和两种酚/氯仿抽提法对卤虫基因组DNA进行了提取;并用紫外光分光光度计法、琼脂糖电泳法和PCR对所提取的DNA进行检测,将它们在DNA的产量、质量等方面的优缺点进行比较,通过三种方法的比较,认为改进的酚-氯仿法是卤虫基因组DNA的最佳提取方法.
The sodium chloride high salt precipitation method and two kinds of phenol-chloroform method were used to extract genomic DNA from Artemia. The resulted DNA samples of these methods were tested by using the UV-visibel spectrophotometer and agarese gel electrophoresis. The difference between yield and quality of the obtained DNA were compared, considering modified phenol-chloroform method as the best.
