纤维素酶产生菌的筛选鉴定和产酶条件优化 被引量:10

摘要 筛选得到1株纤维素酶产生菌TC1,根据真菌的分类鉴定方法,鉴定为根霉属菌(Rhizopus sp.)。正交实验确定该菌株的优化培养条件为:稻草采用15%的醋酸预处理,稻草与麸皮比为3:7,固液比为1:3,接种量10%,温度28℃,稀释1倍的Mandels营养液作为培养基液体。在优化培养条件下培养96h,发酵终点时滤纸酶活FPA、CMC酶活分别达到25.5、249.0 U/g(干物质),较优化前的17.8、187.0 U/g(干物质)分别提高了43%和33%。 One cellulase-production strain TC1 was screened. It was identified as Rhizopus sp. based on identification method of fungi. The optimized fermentation conditions determined by orthogonal design were: straw powder(pretreated by 15% acetic acid), straw powder/wheat bran(3 : 7), solid substrate/liquid nutriment (1 : 3), and quantity of inoculation(10%), 28℃, and half time diluted Mandels was used as liquid nutriment. The maximum activities of filter paper enzyme(FPA) and carboxymethyl cellulose enzyme(CMC) were 25. 5,249. 0 U/g dry medium, respectively under the optimized fermentation conditions after 96h, which were increased by 43% and 33% , respectively compared with the initial one.
