鲁塞尔蝰蛇毒X对血凝学相关蛋白作用的研究

摘要 目的研究鲁塞尔蝰蛇毒 X(Russel viper venomX,RVVX)对凝血相关蛋白的影响。方法将稀释后的蛋白纯品分为对照组和 RVV-X 作用组,同时在有生理性激活剂和无生理性激活剂的情况下,用发色底物法,观察 RVV-X 对凝血因子Ⅶ、Ⅸ、Ⅹ及抗凝血酶,纤溶酶原的活化作用。结果 RVV-X 作用组凝血因子Ⅶ、Ⅸ及纤溶酶原使底物显色微弱增强,与对照组(未加RVV-X)比较,差异有统计学意义(均P<0.05)。凝血因子Ⅹ显色增强明显(P=O.002);抗凝血酶组显色没有变化(P=0.579)。结论 RVV-Ⅹ对凝血因子Ⅶ、Ⅸ及纤溶酶原有弱的激活效果,对于凝血因子Ⅹ是强激活剂,对于抗凝血酶没有激活作用。 Objective To study the effects of russel viper venom Ⅹ (RVV Ⅹ ) on blood coagulation protein. Methods We divide diluted protein into control and RVV-Ⅹ groups, then use chromogenic substract assay to detect the activation effect of RVV- Ⅹ on coagulation factor Ⅶ, Ⅸ, Ⅹ and antithrombin, plasminogen, with or without activator. Results In RVV- Ⅹ group, the coagulation factor Ⅶ, Ⅸ and plasminogen displayed weakly enhanced chromogenesis, all P 〈 0. 05, while the factor Ⅹ group ( P = 0. 002) gave strong chromogenesis. There was no change in of antithrombin group (P = 0. 579 ). Conclusions RVV-Ⅹ has weakly activation effect on coagulation factor Ⅶ, Ⅸ and plasminogen, but strong effect on factor Ⅹ. It has no activation effect on antithrombin.
