摘要 利用RNA i技术,通过干扰秀丽线虫中的sod-3基因表达,建立了因超氧化物歧化酶3(SOD3)表达缺失引起快速衰老的线虫模型,为sod-3基因功能的研究以及相关疾病的基因治疗等提供动物模型. According to the oxidative damage theory of aging, free radicals, particularly reactive oxygen species (ROS), are responsible for the senescence and many human diseases. ROS are generated during normal metabolic process in all the oxygen-utilizing organisms. They are detoxified by superoxide dismutase. The manganese-containing superoxide dimutase 3 (SOD3) has been shown to play a role in the aging process in Caenorhabditis elegans. In this research, we show that sod-3 RNA interference (RNAi) induces insenecence in Caenorhabditis elegans as expected. This method can be used to understand the process of aging and the mechanism of antioxidance.
