SPF级Wistar大鼠血液学及血生化指标正常值范围探讨 被引量:33

摘要 目的 建立本实验室毒理学安全性评价中大鼠血液学及血生化指标的正常值范围;比较断头采血与麻醉后腹主脉采血对生化指标测定结果的影响。方法 收集本实验室近三年来2~3月龄的正常SPF级Wistar大鼠血液学和血生化指标的数值,按照正态分布原理推断其95%可信度正常值范围,并比较了雌雄性别和两种不同采血方式的结果差异。结果 (1)血液学指标中的WBC、RBC、HGB、PLT计数雌雄间无差异,但白细胞分类雌雄性别有差异;(2)血生化指标检测结果雌雄性别有差异,其中断头处死采血方式中,雌性大鼠血清ALT、AST、TG比雄性大鼠低,雌性大鼠TP、Alb、TCH比雄性大鼠高,有统计学意义(P〈0.05);戊巴比妥钠麻醉后腹主脉采血方式中,雌性大鼠ALT、GLU、TG比雄性大鼠低,雌性大鼠AST、TP、Alb、BUN、TcH雄性大鼠高,有统计学意义(P〈0.05);(3)两种采血方式对生化指标有一定影响,断头采血的ALT、AST、TP、Alb、BUN、TCH、TG值较戊巴比妥钠麻醉后腹主动脉采血的测定值高,有统计学意义(P〈0.01),而断头采血的GLU、CRE值较戊巴比妥钠麻醉后腹主动脉采血的测定值低,有统计学意义(P〈0.01)。确立了本实验室雌雄性别Wistar大鼠血常规、血生化正常值范围。 Objective To determine normal ranges of blood and biochemical indexes in laboratory rats for toxicological safety evaluation, and compare the influence of blood extraction by decapitation and by abdominal extraction after anaesthesia on biochemical indexes tests. Method Determine the normal range with 95 % confidence level through normal distribution by processing statistics of all blood and biochemical indexes in 2 - 3 month-old SPF Wistar rats collected over recent 3 years by our laboratory; compare the differences of results from male and female rats and two extraction methods, blood indices. Result (1)There is no difference in WBC, RBC, HGB and PLT counts between males and females, but there is difference in WBC classification between them. (2)Biochemical indexes showed difference between males and females. In results of tests of blood by decapitation ALT, AST, TG in females are lower than in males, but their TP, Alb, TCH are higher than males. Therefore there is statistical significance( P 〈 0.05). In results of tests of abdominal blood, ALT, GLU, TG in females are lower than in males and their AST, TP, Alb, BUN, TCH are higher than males and there is statistical significance ( P 〈 0.05). (3) Methods for blood extraction do have influence on biochemical test results. ALT, AST, TP, Alb, BUN, TCH, TG levels from tests of blood from decapitation are higher than from tests of blood from abdomen after pentobarbital sodium anaesthesia and there is statistical significance ( P 〈 0.01 ) ; GLU, CRE levels from test of blood through decapitation are lower than from the latter method and showed statistical significance( P 〈 0.01 ). Conclusion The normal ranges of blood and biochemical indexes for Wistar rats were therefore established.
