Effect of Bcl-2 loop domain＇s on Bcl-2 construction

目的探讨Bcl-2 Loop domain环状结构域安全检查突变对Bcl-2蛋白结构的影响。方法在同济大学附属东方医院细胞治疗室分别对Bcl-2 Loop domain结构域的55和70位氨基酸行点突变及对Bcl-2 Loop domain结构域行缺失突变并应用PCR及Western blot方法检测Bcl-2 Loop domain结构域突变前后Bcl-2蛋白的表达变化。结果对Bcl-2 Loop domain结构域的55和70位氨基酸进行点突变组，Bcl-2蛋白在上清中和沉淀中均能检测到，对Bcl-2 Loop domain结构域行缺失突变组Bcl-2蛋白只在沉淀中表达，在上清中未检测到。结论Bcl-2 Loop domain结构域在Bcl-2蛋白结构中起到重要作用。

Objective To investigate the effect of Bcl-2 Loop domain mutation on Bcl-2 construction. Method After gene point mutation and deletion mutation on loop domain, expression of Bcl-2 was detected by PCR and Western blot in cell treatment department of East Hospital Affiliated with Tongji University. Results For group which fifty-five and seventy amino acid were mutated, Bcl-2 protein expressed both in the deposition and above liquid. For group which loop domain was mutated by deletion mutation, Bcl-2 protein was detected only in the deposition. Conclusions Bcl-2 Loop domain may play a key role in Bcl-2 protein construction.
