姜黄素及OX-LDL对系膜细胞凋亡及FAP-1和bag-1表达的影响 被引量:6

Effects of Curcumine and OX-LDL on Rat Mesangial Cells Apoptosis and Expressions of FAP-1 and Bag-1 in vitro

摘要 目的:观察姜黄素和氧化低密度脂蛋白(OX-LDL)对大鼠系膜细胞凋亡的作用及其对FAP-1和bag-1表达的影响。方法:采用不同浓度姜黄素和OX-LDL处理体外培养的大鼠系膜细胞后,通过流式细胞仪测系膜细胞凋亡,并采用Western-Bloting检测FAP-1和bag-1表达。结果:OX-LDL能刺激系膜细胞增殖并轻度诱导其凋亡,而姜黄素则显著促进大鼠系膜细胞凋亡,抑制OX-LDL的促增殖作用;OX-LDL对FAP-1表达有轻度诱导作用,而姜黄素显著诱导FAP-1和bag-1表达。结论:姜黄素能显著诱导系膜细胞FAP-1和bag-1表达,通过促进细胞凋亡途径抑制OX-LDL促进系膜细胞增殖作用。 Objective: To explore the effect of curcumine and OX-LDL on rat mesangial cells (MCs) apoptosis and the expressions of Fas-associated phosphatase-1 (FAP-1) and bag-1 in vitro. Methods:Flow cytometic analysis and Western-Blotting were employed to determine the rate of MCs apoptosis and the expressions of FAP-1 and bag-1 on MCs after subconfluent quiescent MCs cultured in vitro were incubated with various concentrations of curcumine and (or) OX-LDL, respectively. Results: OX-LDL promoted MCs proliferation in vitro and induced MCs apoptosis slightly, but curcumine inhibited the proliferation induced by OX-LDL, and promoted MCs apoptosis. OX-LDL increased the expression of FAP-1 slightly, but cureumine promoted the expressions of FAP-1 and bag-1 on MCs significantly. Conclusion: Cureumine can increase the expressions of FAP-1 and bag-l, and inhibit OX-LDL-induced MCs proliferation by inducing apoptosis.
