摘要
目的:研究人工合成的基质金属蛋白酶抑制剂(GM6001)在治疗兔角膜新生血管中的作用。方法:将45只健康新西兰大白兔随机取5只,为正常对照组,余40只作角膜缝线,建立角膜新生血管动物模型,随机分为阳性对照组和200μg/ml的GM6001用药组。裂隙灯显微镜下观察、测量各组新生血管的长度和范围,计算其生长面积,并在造模后不同时间作角膜光镜、电镜观察。结果:在术后不同时间点,GM6001用药组新生血管生长面积较阳性对照组明显减少,差异有显著意义(P<0.001)。组织病理学显示:①阳性对照组可见大量炎性细胞浸润、角膜新生血管丰富。②胶原纤维粗细不等、排列紊乱,而GM6001用药组的上述改变明显减轻,且角膜结构基本接近正常。结论:GM6001对角膜新血管有一定的疗效,降低角膜胶原纤维的破坏及角膜组织中炎性细胞的浸润。
Objective. To study the effect of inhibitor of matrix metalloproteinase GM6001 on the corneal neovascularization. Methods: 5 of 45 rabbits were randomly selected as control group, 40 of 45 rabbits were established as model of corneal neovascularization by suturing in cornea, then randomly divided into positive control group and GM6001 group. The length , extent and size of new vessels in cornea were measured with slit lamp and observed by microscope and electromicroscope. Results: The neovascular size in GM6001 group decreased compared with positive control group, the difference was significant ( P 〈 0. 001 ). The corneal histopathological results showed massive infiltrated inflammatory cells and abundant new blood vessels, disordered collagen fibers in different extent. In GM6001 group, the corneal structure showed less changes and near normals. Conclusion: GM6001 is effective in inhibiting corneal neovasculization and decreasing the infiltrated collagen fibers and inflammated cells.
