摘要
目的观察四氯化碳中毒时神经生长因子(NGF)及其受体P75在大鼠肝组织的表达变化,探讨NGF在肝纤维化发生机制中的可能作用。方法用四氯化碳致大鼠肝中毒模型,用RT-PCR法和Western blot法,分别检测NGF及其受体P75在大鼠肝组织中的表达。结果与对照组大鼠肝组织中NGFmRNA及其受体P75蛋白表达量比较,造模组24 h显著增高(P<0.01,n=6),造模48 h和72 h迅速回降,但仍显著高于对照组(P<0.05,n=6)。结论在四氯化碳中毒性大鼠的肝组织中,NGF及其受体P75的表达可能与肝纤维化的形成密切相关。
Objective By observing the changes in NGF and its receptor P75 in liver of CCl4-induced toxic rats and to evaluate the role of NGF in the mechanism of hepatic fibrosis. Methods The expressions of NGF mRNA and its receptor P75 were detecteded by both hemi-quantitative RT-PCR and Western blot analysis. Results The expressions of NGF mRNA and its receptor P75 in liver of CCl4-induced toxic rats at 24th hour group were higher than that in control group(P 〈0. 01 ,n =6). Then it got down at 48th hour and 72th hour. But it was still higher than that of control group(P 〈 0. 05, n = 6). Conclusion In CCl4-induced rat liver tissue, the expressions of NGF and its receptor P75 is potentially related to hepatic fibrosis.
