摘要
为探讨凹耳蛙Rana tormotus的分类地位,测定了凹耳蛙线粒体12S rRNA基因序列,并从GenBank中下载了蛙科33种蛙的同源序列进行分析,用BI法、NJ法和MP法构建分子系统树。结果表明:3种树拓扑结构基本一致,都显示凹耳蛙与长吻胡湍蛙Huia nasica亲缘关系最近,凹耳蛙与长吻胡湍蛙相聚后再与臭蛙Odorrana组成单系群,而与湍蛙Amolops亲缘关系较远。据此,支持将该蛙从湍蛙属划出,并建议归入臭蛙属。
To study the taxonomic status of the concave-eared frog, Rana tormotus, the 12S rRNA gene sequence of R. tormotus was sequenced and compared with the homologous sequences of 13 Amolops species, 6 Odorrana species, 3 Meristogenys species, 4 Huia species and 4 Rana species retrieved from GenBank. The phylogenetic relationships were reconstructed based on 391 bps using Bayesian Inference (BI), Neighbor-Joining (NJ), and Maximum-Parsimony (MP) method. The topologies of three trees are similar with minor differences. All trees revealed that R. tormotus is most dosely related to Huia nasica. R. tormotus, H.nasica and 6 Odorrana species duster together and form a monophylefic group. The results of this paper support that R. tormotus does not belong to the genus Amolops. We suggest that R. tormotus should be placed in the genus Odorrana.
