钉螺酚氧化酶活性及丙烯基硫脲对其影响的初步观察 被引量:1

摘要 目的:了解钉螺体内酚氧化酶的活性及观察丙烯基硫脲对其活性的影响。方法:①酶组织化学法检测钉螺体内酚氧化酶:采自芜湖江滩的钉螺成螺(包括雄螺和雌螺),解剖后,软体组织置含50 mmol/L邻苯二酚的磷酸盐缓冲液(pH6.8)中。37℃孵育,于第0、5、30 min,体视显微镜下观察酚氧化酶在螺体内的分布并记录。②丙烯基硫脲对酚氧化酶的抑制作用:解剖后的钉螺成螺软体组织,在置入邻苯二酚缓冲液的第0、5、10 min后与丙烯基硫脲作用5 min,最后置入邻苯二酚缓冲液中,观察酚氧化酶催化活性。结果:酶组织化学法显示,钉螺的肝脏部位出现酚氧化酶催化底物邻苯二酚生成的最终产物黑色素。经丙烯基硫脲作用后,酚氧化酶活性被抑制,停止生成黑色素。结论:雌雄钉螺体内均存在酚氧化酶,而且在肝脏部位可能具有较高的酶活性。丙烯基硫脲对酚氧化酶具抑制作用。 Objective:To observe the activity of phenol oxidase (PO) from Oncomellania hupensis and the effect of allyl-thiourea on the enzyme activity. Methods : Anatomized adult female and male Oncomellania hupensis were incubated into pH6.8 phosphate buffer containing 50 mmol/L catechol at 37 12. Some of those snails were observed and recorded for the histological distribution of PO under a stereo microscope. Other snails taken into eatechol for 0,5 or 10 min, respectively, were treated by allyl-thiourea for 5 rain and incubated in eatechol at 37 ℃ for observing and recording the histological distribution of the PO. Results : The results showed that melanin, the product of catechol being treated by PO, appeared on the livers of the snails. However,no melanin produced when the snails were treated by allyl-thiourea. Conclusion :The findings show that PO exists in the body of Oncomelania hupensis and generally locates in its liver. Besides,allyl-thiourea can exclusively inhibit the enzyme activity.
