摘要
采用毛细管电泳法拆分苯磺酸氨氯地平手性对映体并检测其光学纯度,选择CM-β-CD为手性选择剂,考察了CM-β-CD浓度和缓冲液pH值对手性分离的影响,并对分离条件进行了优化。实验结果表明,在不添加任何有机改性剂的条件下,含10 mmol/L CM-β-CD的50 mmol/Ltris-磷酸缓冲液(pH值为2.5)可使对映体在短时间(7.22 min)内完成基线分离,分离度(RS)达4.3。
A capillary electrophoresis method is adopted for chiral separation and optical purity analysis of amlodipine. CM-β-CD is selected as a chiral selector. The influences of concentration of CM-β-CD and pH value of buffer are considered and the separation conditions are optimized. The running buffer is 10 mmol/L carboxymethylated-β-cyclodextrin in 50 mmol/L trisphosphate(pH=2.5) ,which makes amlodipine resolved in 7. 22 min with no organic modifiers and Rs is 4.3.
