摘要
目的研究C-ski和Smad3在大鼠皮肤成纤维细胞辐射性损伤中的变化和意义。方法软X线照射诱导大鼠皮肤成纤维细胞辐射性损伤,Western blot和RT-PCR观察C-ski、Smad3蛋白和mRNA水平的变化,EMSA检测Smad3转录活性的变化。结果辐射后2h,C-ski、Smad3蛋白和mRNA水平、Smad3转录活性无显著变化,4h开始,C-ski表达逐渐下降,Smad3表达和转录活性逐渐升高,在8h分别达到峰谷和峰顶。结论C-ski和Smad3参与大鼠皮肤成纤维细胞辐射性损伤的病理过程,辐射后C-ski下降,Smad3升高,并导致Smad3转录活性增强,细胞增殖受抑,凋亡增加。
Objective To clarify the changes and significance of C-ski and Smad3 in rat skin fibroblast exposed to radiation. Methods The radiation induced injury of rat skin fibroblast was made by exposure to Soft X ray. Protein and mRNA expressions of C-ski and Smad3 were detected by Western blot and RT-PCR. The transcriptional activity of Smad3 was observed by electrophoretic mobility shift assay (EMSA). Results At 2 h after radiation, there were no obvious changes in protein and mRNA expression levels of C-ski, Smad3 or the transcriptional activity of Smad3. At 4 h, the expression of C-ski began to gradually decrease, reaching the minimum at 8 h, and Smad3 and Smad3 transcriptional activity began to increase, peaking at 8 h. Conclusion C-ski and Smad3 are involved in the pathological process of radiation induced fibroblast injury. Radiation can decrease C-ski and increase Smad3 expression, resulting in elevated Smad3 transcription activity, therefore cell proliferation is repressed and cell apoptosis is increased.
